For me, it's usually either the "quote" when at Lilac heist, River points to her right, Zoe raises her eyebrows in doubt, and River replies with her "yeah, dummy" look. That, or when Kaylee is saying godbye to Simon with the Capisan 38 Engines just falling out of the sky - which Joss also said was his favorite delivery of a line in the film.
I fall else fails, "My Turn" ought to get ya.

Operative: "Its worse than you know..." Mal: "It usually is."

I still love and always will the quote I have at the bottom of my post;
"Its worse than you know"
This appies to almost everything in my life, and other people lives too I hear.
and I do like
"except for being still crazy..." applies to all the people I work with!!!!!

I think one of my all time favorite parts in Firefly is in the “Jaynestown” episode when Jayne confesses to his admiring masses on Canton that he isn’t a hero, that in reality he is nothing but a two bit crook. That scene is just powerful to me and has always been a favorite.

Jayne had gotten blown away by all the hype and honor thrust upon him by his adoring townspeople. It was like everything he’d done selfishly wrong in the past had been wiped away and he was ready to start over. Nobody knew the real Jayne. From there he could be a success, write his family and tell them he was a respected citizen. He could be like Mal, maybe better. Deep down he knew it would never happen but for some reason it was his fantasy. For a brief time he could be KING.

Adam Baldwin did one of the best acting scenes of the series right then. After that the Whedon verse had it’s hooks in me. Then in the scene in the cargo bay where Jayne is talking to Mal we see that although Jayne didn’t understand, the mudders had forgiven him for his past. All they knew was that he had dropped a box of money on them, at a time when it was probably needed the most. That made him their hero no matter what had happened before.

Usually if I’m lumbering around the house or happen to think of Firefly someplace, that’s the scenes that comes to mind.

SERENITY (BDM) QUOTES

Wash: This landing is gonna get pretty interesting.
Mal: Define “interesting”.
Wash: Oh God, oh God, we’re all going to die?

Mal: This is the captain. We have a little problem with our entry sequence, so we may experience some slight turbulence and then - explode.

Zoë: At last, we can retire and give up this life of crime.

Mal: We’re coming down to empty the vault now!
Vault Guard: You’ll have to give me your authorization password!
Jayne fires a burst from his automatic rifle
Vault Guard: …Okay!

Jayne: Boy, it sure would be nice if we had some grenades, don’t you think?

Mal: Faster! Faster! Faster would be better!

Kaylee: Goin’ on a year now I ain’t had nothin’ twixt my nethers weren’t run on batteries!
Mal: Oh, God! I can’t know that!
Jayne: I could stand to hear a little more.

Jayne: My muscular buttocks it’s forty.

Fruity Oaty Bar Jingle: Fruity Oaty Bars! Make a man out of a mouse! Fruity Oaty Bars! Make you bust out of your blouse! Eat them all the time! Let them blow your mind… ohh! Fruity Oaty Bars!

Fanty: Do you know that girl?
Mal: I really don’t.

Simon: Eta Kooram Nah Smech!

River: Miranda…
Mal: Miranda? Go back further.
Mr. Universe: No…
Mal: Uh… please?
Mr. Universe: Oh, Mal, you’re very smart. Someone is talking to her.
Wash: The oaty bar?

Mal: Do you want to run this ship?
Jayne: Yes!
Mal: Well… you can’t…

Mal: Dear Buddha, please bring me a pony and a little plastic rocket…

Mal: Y’all got on this boat for different reasons, but y’all come to the same place. So now I’m asking more of you than I have before. Maybe all. Sure as I know anything, I know this - they will try again. Maybe on another world, maybe on this very ground swept clean. A year from now, ten? They’ll swing back to the belief that they can make people… better. And I do not hold to that. So no more runnin’. I aim to misbehave.

FIREFLY151

For me it's when Mal & River are piloting Serenity at the end of the movie, and they pass through the storm...
And lots of times in the music, especially 'Serenity' from the movie soundtrack.

Before I post some of my fav scenes and quotes I gotta say it Z. Your description of AB's acting in Jaynestown (one of my favs in the series) is spot on.

If you weren't hooked already, Jayne's concern over whether they should use his new-found fame to do the "job" is classic. The "Me, Jayne Cobb" line, and Mal's response, also classic. He probably thought that he would never be capable of doing any good, and here this town puts him on a pedestal, literally.

"You had a riot on account of me?"

Okay, the series:

When I thought it was something special -

1. The moment I saw Serenity flying over the horses in the original promos.

2. In the pilot, no words could do this scene justice: Jayne kneeling by the infirmary as Simon tends to Kaylee.

3. Pilot: Mal pauses for a milisecond just before he "pushes" Inara toward her shuttle while being chased by the Reavers. Right there you know something is up with these two.

4. My old signature: Again, the pilot, Wash's "Here's something you can't do." In that moment you know both he and Serenity are something special.

5. Finally, again Pilot, Kaylee's "That's my girl"
Right there Serenity is not just a ship, she's another character, because Kaylee said so.

Fell in, no turning back, love:

OoG - we got a taste of the crew's backstories, enough to whet our appetite and have us begging for more.

Signed, sealed, delivered - War Stories.

OMR - when Zoe is teasing Mal about his marriage to Saffron. Book's look when he says "Special Hell"

The BDM:

Here's something I take note of each time I see it. Mal argues with Simon and sarcastically tells him "there may be tears" in regards to protecting his sister. But when the Reavers interupt the heist, he grabs River and makes sure she gets on the Mule. And later in the Maidenhead he steps over Jayne, scoops her up and carries her off. His actions almost always belies the cynic in him. Just an observation. Love it.

River's look at Zoe during the Heist (Summer was so good here, no wonder they cast her for SCC).
The Maidenhead Bar - one of my fav lines "Twixt my nethers" and Mal's "Oh God, I can't know that." Joss does so much with just a few lines. Kaylee is like a sister to Mal, and do you really want to know about your sister's (ahem) "nether region."

One final thought in the BDM: After Mal finds Mr. Universe sprawled out by his love-bot, his recording tells Mal it's hard to get to. He goes to send out the signal, and when he comes upon the canyon that is the Signal room he says "Hard to get to!" Great line, delivered perfectly by Nathan to show the irony of Mr. U's understatement.

But perhaps my most fav line of all time is River's "My turn."

The more I think about it, the more I love this movie.
